Silodosin works by relaxing smooth muscle in the prostate and bladder neck, helping urine flow more easily. A key caution is that it may cause dizziness or low blood pressure, especially when standing up, so care is needed when driving or getting up quickly.
Indications
- Treatment of signs and symptoms of benign prostatic hyperplasia in adult men.
- It is not used to treat prostate cancer and does not shrink the prostate permanently.
Dosage & Administration
For adults, Silodosin is usually taken once daily with food, at the same time each day. The exact dose depends on the prescribed strength, kidney function, other medicines, and tolerability. Do not start, stop, or change the dose without the prescribing doctor's advice.
Side Effects
- Common: dizziness, light-headedness, headache, diarrhoea, nasal congestion, and reduced or absent ejaculation.
- May cause low blood pressure on standing, especially after the first doses or with blood pressure medicines.
- Serious but uncommon: fainting, allergic reaction such as swelling or breathing difficulty, and prolonged painful erection.
- Can cause intraoperative floppy iris syndrome during cataract or glaucoma surgery.
Precautions & Warnings
Use caution in people with low blood pressure, a history of fainting, kidney disease, liver disease, or those taking medicines for high blood pressure or erectile dysfunction. Tell the eye surgeon before cataract or glaucoma surgery if Silodosin is being used or was used in the past. Use this medicine under the guidance of a registered doctor or pharmacist.
Drug Interactions
- Strong CYP3A4 inhibitors such as ketoconazole, itraconazole, clarithromycin, ritonavir, or similar medicines can greatly increase Silodosin levels and should generally be avoided or are contraindicated depending on local guidance.
- Other alpha-blockers may increase the risk of low blood pressure and are usually not combined.
- Blood pressure-lowering medicines and nitrates may increase dizziness or fainting risk.
- PDE5 inhibitors such as sildenafil, tadalafil, or vardenafil may increase the risk of symptomatic low blood pressure.
- Potent P-glycoprotein inhibitors may increase exposure; medical advice is needed.
Contraindications
- Known hypersensitivity to Silodosin or any component of the capsule.
- Severe kidney impairment.
- Severe liver impairment.
- Concomitant use with strong CYP3A4 inhibitors, where product guidance lists this as contraindicated.
Pregnancy & Lactation
Silodosin is intended for use in adult men with benign prostatic hyperplasia and is not indicated for women. Safety in pregnancy or breastfeeding has not been established; pregnant or breastfeeding women should not use it unless a specialist specifically advises otherwise.
